Cinco años después del éxito de "La gran aventura de Mortadelo y Filemón", llega una nueva entrega de las peripecias de los disparatados agentes creados por Ibáñez, esta vez con la dirección de Miguel Bardem. El planeta se enfrenta a una gran amenaza. Coincidiendo con la peor sequía que se ha vivido, un grupo de malhechores está terminando con todas las reservas de agua que quedan en la Tierra. Los mejores agentes secretos del mundo se reúnen urgentemente, pero ni siquiera cuentan con una pista. Para acabar de empeorar las cosas, el malvado Botijola planea atentar contra esa reunión. Su intención no es otra que convertir la bebida que lleva su nombre en el sustituto del agua. Para ello, intentará también secuestrar al alocado profesor Bacterio y conseguir que trabaje para él. Sin embargo, hay dos cosas con las que Botijola no contaba. Por un lado, Bacterio, antes de ser secuestrado, escondió las piezas de su invento en distintos momentos históricos, utilizando la máquina del tiempo. Por otro lado, hay dos de los agentes internacionales que no han acudido a la reunión saboteada, dos agentes que van a convertirse en la única esperanza del mundo: Mortadelo y Filemón.

Life is uncomplicated and safe, and although Pencil is a Beagle, Peggy is happily entrenched in a satisfying relationship of co-dependency with her beloved pet. This stable status quo is rocked when Pencil’s sudden death leaves Peggy reeling in a world devoid of meaning without him. Her quest for happiness leads her to the attractive, yet ambiguous Newt (Peter Sarsgaard) who introduces her to animal activism, a vegan lifestyle and even a new dog. For photographer Ben (Joshua Jackson) and his new wife Jane (Rachael Taylor), his new assignment - a lucrative fashion shoot in Tokyo - was supposed to be a kind of working honeymoon. With this exotic professional opportunity and the limitless possibilities of a new marriage, Ben and Jane arrive in Japan. But as they make their way on a mountain road leading to Mt. Fuji, their new life together comes to, literally, a crashing halt. Their car smashes into a woman standing in the middle of the road, who has materialized out of nowhere. Upon regaining consciousness after the accident, Ben and Jane cannot find any trace of the girl Jane believes she hit with the car. Shaken by the accident and by the girl's disappearance, Ben and Jane arrive in Tokyo, where Ben begins his glamorous assignment. Having worked in Japan before and fluent in the language, Ben is comfortable there, and he eagerly reunites with old friends and colleagues. Jane, a newcomer to the city, feels very much like a stranger in a strange land as she makes tentative, unsettling forays through the city. Ben, meanwhile, has discovered mysterious white blurs - eerily evocative of a human form - that have materialized on an entire day's work from the expensive photo shoot. Jane's concerns escalate as she believes the blurs in Ben's photos are the dead girl from the road, who is now seeking vengeance for them leaving her to die.
